Description

Iclusig (Ponatinib) tablet is used in the treatment of adults with leukaemia (cancer of

the white blood cells). It is used to treat the different stages of chronic myeloid

leukaemia (CML), namely chronic, accelerated and blast phases. It is also used in the

treatment of acute lymphoblastic leukaemia (ALL) in certain patients who are

‘Philadelphia-chromosome positive’, which means those patients whose genes have

rearranged themselves to form another special type of a chromosome called

‘Philadelphia-chromosome’. This chromosome leads to the development of Leukaemia.

Pharmacodynamics

Ponatinib is a multi-target kinase inhibitor. Its primary cellular target is the Bcr-Abl

tyrosine kinase protein which is active and works to promote the progression of CML.

This protein arises from the fused Bcr and Abl gene which is commonly known as the

Philadelphia chromosome. Ponatinib is especially useful in the treatment of resistant

CML because it works to inhibit the tyrosine kinase activity of Abl and T315I mutant

kinases. The T315I mutation highly confers resistance in cells. It prevents other Bcr-Abl

inhibitors from binding to the Abl kinase. Some other targets that ponatinib inhibits are

members of the VEGFR, PDGFR, FGFR, EPH receptors and SRC families of kinases, and

KIT, RET, TIE2, and FLT3.

Contraindications

This medicine is not indicated to the patients who have a known allergy to any of the

ingredients of this medicine.

Dosage

Normal Dose

Missed Dose

When you miss a dose, make sure to tell your healthcare provider regarding the same to

get proper guidance. Do not try to double the dose strength in order to catch up.

Usage Of Medicine

Iclusig (Ponatinib) tablet is used in the treatment of:

● chronic myeloid leukaemia, and

● acute lymphoblastic leukaemia.

Warning And Precautions

decrease the dose strength as per your own wish. Do not crush, chew or break

the tablet.

● Do tell your healthcare provider beforehand about your medical condition and

medical history, along with the list of medicines you already take.

● In patients with kidney disease, administer this medicine with utmost caution.

● In patients with liver disease, administer this medicine with utmost caution.

● During pregnancy/breastfeeding, take it only if your healthcare provider

prescribes it. Make sure to talk about the potential risks and benefits.
